The most common way is using 一番 (いちばん) with a negative predicate, or the pattern 最も〜ない (もっとも〜ない). Other patterns include 最小限 (さいしょうげん) for 'at least' in a minimal sense, and 少なくとも (すくなくとも) for 'at the very least'.

一番少ない only works for countable quantities or measurable amounts. For abstract qualities like 'least interesting', use 一番面白くない, not 一番少ない面白い.
